Spider-Man vs. Venom is a soft-reboot of the Spider-Man film series and the first installment of a shared universe based on characters that appear in Spider-Man publications from Marvel Comics. The film was directed by Drew Goddard who co-wrote the script with Dante Harper.

- Taron Egerton as Peter Parker/Spider-Man: A brilliant Teenager and photographer for the Daily Bugle who gets spider-like abilities from a radioactive spider and uses them to take up the persona of a spider-based vigilante protecting New York City from its criminal underworld.
- Justin Prentice as Eddie Brock/Venom: A young reporter for The Daily Bugle Gwen's former boyfriend he becomes jealous of Peter and then becomes the archenemy of Spider-Man Venom.
- Cameron Monaghan as Cletus Casady/Carnage: A violent, murderous psychopath who bonds with the carnage symbiote, becoming a common threat to both Spider-Man and Venom, ultimately forcing them to put their differences aside and team up to stop him.
- Britt Robertson as Gwen Stacy: Peter's girlfriend and next-door neighbor. She is the first person Peter reveals his identity to, and the one who often fixes his costume and treats his injuries.
- Meryl Streep as May Parker: Peter's aunt whose she raised with her husband, Ben Parker. She regularly sees a therapist to deal with the death of Ben while she's trying to pay his nephew's studies.
- Nat Wolff as Harry Osborn: Peter's best friend and ally
